Animal experiments confirm the effectiveness of trispecific antibodies

Scientists at the Vaccine Research Center used monkeys and two SHIV strains as models to test for trispecific antibodies. One of the SHIVs was sensitive to the neutralization reaction induced by VRC01 and the trispecific antibody, but did not respond to the neutralization of PGDM1400. Another type of SHIV is sensitive to the neutralization of PGDM1400 and trispecific antibodies, but is not sensitive to the neutralization of VRC01.

They selected 24 monkeys and divided them into 3 groups to inoculate VRC01, PGDM1400 and trispecific antibodies. Five days later, the researchers gave the monkeys a SHIV virus. The results showed that 5 monkeys vaccinated with PGDM1400 antibody and 6 monkeys vaccinated with VRC01 antibody were eventually infected with SHIV. However, monkeys vaccinated with trispecific antibodies were not infected with HIV. This suggests that this three-in-one antibody is effective in preventing HIV.

Sanofi is producing trispecific antibodies for clinical phase I, and trials are expected to be conducted by the end of 2018 to test antibody safety and pharmacokinetics. In addition, the NIAID-funded AIDS clinical trial team is planning to conduct a separate clinical Phase I trial.

Mascola said: "The cooperation between NIAID and Sanofi has a milestone. We will try to push this specific antibody from the laboratory to the clinic."
